注意:
1.所有溶剂和缓冲液应脱气本协议是为制备二磺基 - Cy5标记的IgG抗体而设计的。它被设计为标记1毫克。这假定平均蛋白质分子量为155K道尔顿。
1.将抗体溶解在偶联缓冲液(10mg / ml)中。将1mg要标记的抗体(0.1ml)置于0.5ml微量离心管中,并用氮气冲洗。
2.将150μl高碘酸钠溶液(0.1 M)加入抗体中,密封并在环境温度下无光条件下充分混合30-45分钟。
3.当氧化反应正在孵育时,将凝胶过滤柱平衡到偶联缓冲液中。将高碘酸盐反应物置于柱上:收集并合并含有抗体的级分
1.将二磺基-Cy5酰肼溶解在偶联缓冲液中,得到0.02M溶液。
2.将等体积的抗体与二磺基 - Cy5酰肼溶液混合并在环境温度下轻轻混合3小时或在4℃下过夜。
3.使用已经用PBS预处理的10ml凝胶过滤柱来纯化产物。用PBS洗脱产品。
4.分离个有色级分,并从PBS空白样品中扫描250-800nm的样品。
1.未缀合的染料也可以通过透析与标记的抗体分离。透析不能像凝胶过滤那样有效快速分离。建议尽可能使用凝胶过滤。
提供材料干燥的染料标记1mg羰基化合物。
所需材料但未供应偶联缓冲液:1:1甲醇/乙酸三乙铵缓冲液,pH 8.0 Sigma-Aldrich(90358)。
注意:
1. pH会改变反应的标记效率。宜标记发生在pH 7.2。
2.所有溶剂和缓冲液应脱气。该方案用于用Cy5酰肼试剂标记〜7.8μmol的辛醛。
1.在1:1甲醇/ TEAA缓冲液,pH8.0溶液中将二磺基-Cy5酰肼溶液制成1mg / ml并用氮气冲洗。
2.在1:1甲醇/ TEAA缓冲溶液(pH8.0至1mg / ml)中制备羰基衍生物溶液并用氮气冲洗。
3.将两种溶液加在一起并轻轻混合3小时。
4.真空除去反应溶剂。
1.通过反相HPLC纯化产物。在DiSulfo - Cy5 650 nm波长处监测。
