含有荧光报告基团6-氨基-1-萘磺酰胺(ANSN)的底物是监测各种丝氨酸蛋白酶(1-12)酶活性的有用化合物。在这类化合物中,ANSN报告基团(在R1位置)与短的三肽序列相连。肽序列的设计是为了优化酶和底物之间的相互作用。可以添加到R2和R3位置的附加组分反映了P'亚基位置的变化,并且通常会影响底物的动力学参数。作为有效底物的化合物在三肽和ANSN基团之间水解。一旦从肽段分离,ANSN基团的相对荧光强度增加了约1000倍。
下页确定的动力学特性将有助于选择合适的底物。ANSN底物被证明特别适用于因子VIIa(1-5,7,9,10,12)的分析。尽管仅因子VIIa的底物水解速率相对较慢,但当将组织因子纳入分析系统时,化合物SN-17a等几种化合物的kcat发生了很大变化。
使用信息:
在二甲基亚砜中,以10mM的储备溶液形式提供基于ANSN的底物,通常用于分析浓度范围为400 nM至50pM的酶。ANSN底物通常在50至100µM的终浓度下使用,并且可以在大多数生理缓冲液(即TBS、HBS、PBS等)中稀释到该终浓度。反应(荧光的相对变化)可使用标准荧光计或荧光板阅读器进行监测。仪器设置在以下波长(激发=352nm,发射=470nm)时,可以使用动力学或端点模式。通过在发射光束中使用390到450nm长通截止滤波器,可以将光伪影小化。
